It was the son's Y-DNA which was analyzed. Not even mtDNA was recovered from
the fragmentary remains of Columbus himself.
I suspect the technique used for Y-DNA was based on the specialized SNP
techniques developed at Orchid (mentioned in the show). It works on smaller
fragments of DNA.
